Is testng works on non web browser based application(Is that Desktop application). - testng is a unit testing framework. it works on no browser or desktop application. However selenium works on browsers.

If agile is there, how automation starts - i personally dont prefer that since automation is good to do on a stable project. Once you feel that project is stable and there are no functional defects, you can automate

One more thing - by the level of questions - interviewer was not aware of selenium at all :)
